This activity was planned to meet the educational needs of clinical pharmacists across disciplines who shepherd new drug approvals through acquisition, pharmacy and therapeutics bodies and integration into clinical practice

Learning Objectives

After completing this activity, the learner should be able to:

Briefly discuss challenges with laboratory-intensive methods for absorption, distribution metabolism and excretion (ADME) characterization for compound leads in development
Describe physiologically based pharmacokinetic (PBPK) modelling and its key limitations for predicting lead compound pharmacokinetics
Describe how machine learning (ML)-enhanced Quantitative Structure-Activity Relationship (QSAR) predict input parameters that can improve PBPK simulation accuracy (e.g., clearance, partition coefficients, plasma protein binding)
Illustrate ML-enhanced computational approaches applied across drug development stages
Outline Absorption, Distribution, Metabolism, and Excretion (ADME)-derived parameters that can be integrated in model-informed drug development (MIDD) to support regulatory-decision making
Describe three relevant appliactions of MIDD that facilitate regulatory decision-making (e.g., dose selection or estimation, clinical trial simulation, or mechanistic safety evaluation)
